摘要
实验表明人脑胶质瘤原代培养细胞及4个人脑恶性胶质瘤体外细胞系细胞培养上清液(Su-pernatant,SN)显著抑制植物血凝素PHA-P刺激的正常人或胶质瘤患者自体和异体外周血淋巴细胞增殖。我们发现用抗转化生长因子-β_2(TGF-β_2)单克隆抗体可显著降低胶质瘤SN的免疫抑制活性。4个人脑胶质瘤体外细胞系应用抗TGF-β_2单抗免疫组化染色呈阳性反应,3例正常脑组织呈阴性反应。Northern杂交实验表明上述4个人脑胶质瘤体外细胞系均表达6kb的TGF-β_2mRNA,而4例人胎脑则无表达。这些结果提示人脑胶质瘤细胞可以自体分泌免疫抑制因子抑制患者的免疫功能,而这抑制因子的主要成份是TGF-β_2。
The effect of the supernatant (SN) of 7 cultured tuman primary glioma cells & 4 malignant human glioma cell lines on the proliferation of peripheral blood lymphocytes (PBL) from normal healthy subjects,autologous & allogeneic PBL of patients with gliomas activated by mitogen PHAI-p was studied.It was found that all the SN of different types of cultured glioma cells inhibited significantly the mitogenic response of PBL,either from normal controls or from patients with gliomas.This evidence indicated that the human glioma cells secreted an immunosuppressive factor which was autocrine in character.It was reported that the glioblastoma-derived T cell suppressor factor (G-TsF) might be a member of TGF-βfamily identified as TGF-β_2.We found that the effect of the SN on the proliferation of PBL activated by PHA-p was greatly depressed by anti-TGF-β_2 monoclonal antibody. The expression of TGF-β_2 in 4 malignant human glioma cell lines & 3 normal human brain specimens was examined by immunocytochemistry. The result showed that there was a positive expression of TGF-β_2 in 4 malignant human glioma cell lines, while no expression was found in 3 normal adult brain tissues. The gene expression for TGF-β_2 was further investigated by Northern blot hybridization analysis in 4 malignant human glioma cell lines & 4 human fetal brain tissues, Whereas 4 glioblastoma cell lines expressed TGF-β_2 mRNA, but human fetal brain tissues did not express detectable levels of TGF-β_2mRNA. It was suggested that the autocrine of G-TsF/TGF-β_2 by glioma cells in vivo may contribute to the escape of immune surveillance for the development of gliomas.
