摘要
目的观察健忆口服液对老年小鼠脑组织中β淀粉样蛋白(Aβ)和Tau蛋白的作用及其与单胺类神经递质的相关性.方法设昆明种老年小鼠(18～20月龄)为中药治疗组(健忆口服液20g·kg-1·d-1)和对照组;另设昆明种小鼠(8～10月龄)为青年对照组.灌胃给药2～4周.检测:小鼠全脑组织Aβ、Tau蛋白、去甲肾上腺素(NA).结果青年与老年鼠相比全脑组织NA、Aβ和Tau蛋白含量水平差异均有高度显著性(P<0.01);服药2周后,治疗组Aβ含量变化较对照组差异有显著性(P<0.05);疗程4周时,NA、Aβ和Tau蛋白含量变化,在治疗组与对照组之间差异均有高度显著性(P<0.01),但与青年鼠比较差异仍有显著性(P<0.05);老年小鼠脑组织NA含量与Aβ、Tau蛋白含量之间,在对照组和治疗组均无显箸相关性.结论健忆口服液对老年小鼠脑组织Aβ和Tau蛋白具有明显改善治疗作用,提示可能对阿尔茨海默病相关指标具有相似治疗作用;NA与Aβ、Tau蛋白无相关性,不能相互替代检测.
Aim To observe the effect of Jian yi Kou Fu Ye（JYKFY） on amyloid-β-peptide（Aβ） and Tau-protein,and the correlation between the cerebral proteins and Noradrenalinne（NA） in aging mice, discuss its treating action to the patients with Alzheimer＇ s disease （AD）. Methods Aging mice （ 18 -20 months） were randomly assigned to treated JYKFY 20 g·kg^-1·d^-1 and control group,meanwhile young mice （8 - 10 months） as comparison. The Aβ,Tau-protein and NA were examined after 2 and 4 weeks. The differences between groups were compared by T-test, and the correlation analysis was done between Aβ ,Tau-protein and NA. Results The difference of Aβ ,Tau-protein, NA between young and aging mice was very. significant （ P 〈0.01 ） ;after 2 weeks, the change of Aβ in treated group was significant compared to that in control （ P 〈 0.05 ） ; and in the end of 4 weeks, the difference of Aβ,Tau-protein and NA between treated and control group was all very significant （P 〈 0.01 ） ;there was not significant correlation between NA and Aβ,Tau-protein in the treated neither in the control group. Conclusion JYKFY had significant action to ameliorate cerebral Aβ and Tau-protein in aging mice, and it means that it had perhaps equal action on amyloidal plaque （ induced by Aβ） and neurofibrillary lesion （ by Tau-protein） in patients with AD. The examination of Aβ and Tau-protein cannot be remplaced by that of NA.
