黄芪提取物对大鼠全脑缺血再灌注后IL-1β、TNF-α和IL-6表达的影响 被引量:12

摘要 目的研究黄芪提取物(EA)对大鼠全脑缺血再灌注炎症损伤的保护作用。方法采用大鼠四动脉结扎模型,观察EA对大鼠缺血再灌注大鼠神经功能学评分、血清IL-1β、IL-6、TNF-α含量及脑组织IL-1β、TNF-α的含量的影响。结果EA对全脑缺血再灌注24h后的大鼠神经功能有改善作用;EA能降低大鼠全脑缺血再灌注后血清IL-1β、TNF-α、IL-6和脑组织IL1-β、TNF-α的含量。结论EA对大鼠全脑缺血再灌注损伤有一定的保护作用,其机制可能与抑制IL-1β、TNF-α、IL-6的表达有关。 Objective To explore the protective effects of extract of astragalus on inflammatory injury after global cerebral ischemia/reperfusion in rats. Methods The model of global cerebral ischemia/reperfusion was established with four vessel occlusion method(4-VO). Sprague-Daewley rats were randomly divided into therapy groups, shame operation group and ischemia/reperfution group. Vertical screen test and beam balance test were used to evaluate the rats' behavioral function at 12 h and 24 h after reperfusion. After global cerebral ischemia/reperfusion, the rats' serum and cortex IL-1β, IL-6 and TNF-α were detected with radio -immuno-assay. Results Compared with ischemia/reperfusion group, serum and cortex IL-1β, IL-6, TNF-α of EA (40,80 mg/kg) group declined significantly after global cerebral ischemia/reperfusion. Behavioral function of EA group after 24 h reperfusion was improved to some extent. Conclusion EA has protective effects on cerebral ischemia which is related to inhibition of cytokine expression.
