20(S)-人参皂苷Rg_3与化疗药物联合应用对S180肉瘤的影响 被引量:5

摘要 目的:探讨20(S)-人参皂苷Rg3(SPG-Rg3)与化疗药物联合应用对S180肉瘤生长以及荷瘤小鼠生命延长期的影响。方法:体外通过染料排斥实验观察SPG-Rg3抗肿瘤细胞生长的作用。体内建立S180实体瘤模型,将荷瘤小鼠随机分为6组,即SPG-Rg33、1和0.3 mg.kg-1组及环磷酰胺组(CTX)、溶剂对照组(Control)以及联合用药组(CTX+SPG-Rg3),每日腹腔内给药,持续10 d,停药24 h后处死动物,称体重、瘤重,计算肿瘤生长抑制率(%)及两药相互作用指数(CDI)。并建立S180腹水瘤模型,动物随机分为SPG-Rg33、1和0.3 mg.kg-1组及环磷酰胺组(CTX)、溶剂对照组(Control)以及联合用药组(5-FU+SPG-Rg3)6组,连续5 d腹腔给药,记录小鼠存活天数,计算生命延长期。结果:体外实验中,SPG-Rg3抑制了S180肉瘤细胞生长,其IC50值为(10.49±2.96)mg.L-1。体内实验中,SPG-Rg3对S180肉瘤生长有明显的抑制作用,(SPG-Rg3+CTX)联合用药组抑制肿瘤生长的作用优于SPG-Rg3和CTX组。SPG-Rg3和5-FU均延长荷瘤小鼠的生存期,(SPG-Rg3+5-FU)联合用药组生命延长率比5-FU单独用药组明显增高。结论:SPG-Rg3可明显抑制肿瘤的生长,延长荷瘤小鼠的生存期,且SPG-Rg3可以提高CTX和5-FU的抗肿瘤作用,有一定的协同增效作用。 Objective To study the effects of 20(S)-ginsenoside Rga (SPG-Rga) on growth of sarcoma 180 and life of mice with sarcoma 180. Methods In vitro, stain exclusion experiment was used to observe the effect of SPG- Rg3 on sarcoma 180 cells. In vivo, the mice models of sarcoma 180 were built and divided into 6 groups: 3, 1, and 0. 3 mg · kg^-1 SPG-Rg: groups, CTX group, control group, and SPG-Rg3 combined with CTX group. Twenty-four hours after inoculating sarcoma cells, mice were administrated by abdominal cavity and killed at the 10th day. The body and neoplasm weights were detected, and the growth suppression rate of tumor and CDI were calculated. The model of ascites sarcoma 180 was also built and CTX was replaced by 5-FU. The method of diviving groups was same as before. Mice were administrated for 5 d and after that, the life of mice with sarcoma 180 was recorded and life elongation term was calculated. Results In vitro, SPG-Rga inhibited the growth of sarcoma 180 cells and the ICa0 value was (10. 49 ± 2.96) mg · L^-1. In vivo, SPG-Rga also inhibited the growth of sarcoma 180 and the inhibitory effect of SPG-Rga combined with CTX was superior to those of SPG-Rga or CTX. SPG-Rga and 5-FU also prolonged life of mice and the rate of life elongation in SPG-Rgs combined with 5-FU group was higher than that in 5-FU group. Conclusion SPG-Rgs can inhibite the growth of sarcoma 180 and prolong the life of mice with sarcoma 180. Foremore, SPG-Rgs can reinforce the inhibitory effects of CTX and 5-FU on tumor growth.
