摘要
在pH值为7.2,温度为40℃的条件下,As1.398中性蛋白酶水解大豆分离蛋白,采用pH-stat法结合Ex-cel软件方便准确地求出酶促反应的初始速度,从而根据Lineweaver-Burk双倒数方程计算出该酶的最大反应速度(Vmax)和米氏常数(Km).实验条件下,As1.398蛋白酶的Km值为1.46 mg/mL.
The enzymatic hydrolysis of soy protein isolate (SPI) was carried out in a certain condition (pH 7.2, 40 ℃) with AS1. 398 neuter protease. In this course original reaction velocities were deduced with pH stat method combining with Excel software, then according to Lineweaver-Burk dual reciprocal equation, the Vmax and Km were calculated. Km value was 1.46 mg/mL under the experimental condition.
