携带Ang-1的MSCs双介入治疗ANFH的转染与表达 被引量:2

The Transfection and Expression of the Femoral Head on the Double Interventional Therapy of Mesenchymal Stem Cells with Angiopoietin 1 Gene in ANFH

摘要 目的建立兔股骨头坏死模型,探讨双介入治疗方法的可行性,检测治疗后股骨头的转染率及表达。材料与方法用激素建立兔股骨头坏死模型80只,随机分为钻孔注入组、动脉灌注组、双介入组、对照组。实验组采用经皮穿刺于股骨头钻孔,直接注入携带血管生成素-1(Ang-1)的骨髓间质干细胞(MSCs),或/和经颈动脉穿刺,行超选择插管至双侧股骨头供血动脉,缓慢注入携带Ang-1的MSCs,对照组不作治疗。术后2、4、6、8周分别处死动物,取股骨头进行组织学观察、荧光检测、免疫组织化学检测。结果介入组转染增强型绿色荧光蛋白(pEGFP)/Ang-1质粒的MSCs分化的细胞在荧光显微镜下见高表达Ang-1基因,同时局部循环血管和成骨细胞增多。明显好于单独治疗组、对照组。结论携带Ang-1的MSCs经双介入治疗不仅能促进毛细血管的生成,而且能定向进行骨分化。 Objective To establish rabbit model to study the feasibility of the double interventional therapy, and detect the efficiency and expression of the femoral head. Materials and Methods To establish an rabbit model with eorticosteroid induced avascular necrosis of the femoral head. Rabbit models were randomly divided into group with the percutaneous drilling into the femoral head, artery embolization, double interventional therapy and control group, and direcdy administer mesenchymal stem cells with angiopoietin 1 gene expression, and make the carotid puncture to perform the superselective eatheterization to the blood supply vessels of the bilateral femoral heads and slowly administer mesenchymal stem cells with Angiopoietin gene expression. The rabbits in the control group were not treated. 2,4,6,8 weeks after the double (dual) interventional therapy, The samples of the bones and the blood vessels were observed by histology, stained with imnmnohistochemical and with fluorescent. Results In the double (dual) interventional therapy group bright green fluorescence could be watched with Fluorescence Microscopy in pEGFP/ Ang 1 transfected MSCs. Inmmnochemical staining with Ang 1 in engineered cell, which was strongly positive. There was a stable and efficient expression of Angl gene, an increase of microscopic blood vessel anastomosis in the femoral heads after the therapy and there was more new bone formation, which were better than that in the test group alone and in the control group. Conclusion Mesenchymal stem cells with Angiopoietin gene expression can not only enhance the formation of new microscopic blood vessels but also determine osteoblast proliferation and differentiation by treatment of double interventional therapy.
