摘要
目的:研究自体浓集骨髓基质干细胞(MSCs)复合物的成骨作用,以寻求治疗股骨头缺血性坏死的新方法。方法:中国白兔24只,随机分成3组。小牛脱钙松质骨(DBCB)作为载体,将一定量的骨形态发生蛋白(BMPs)与不同浓度含有骨髓基质干细胞(MSCs)的单核细胞悬液复合,制成MSCs/BMP/DBCB复合物(MⅠ:1×106,MⅡ:1×105和MⅢ:1×104),分别回植在骶棘肌内。术后2、4、8、12周取材,对标本做放射影像学、大体观察、显微镜组织学观察。结果:MⅠ组新骨生成量多于其它组。结论:复合物具有治疗股骨头缺血性坏死的潜能,但有待进一步研究。
Objective:To observe the osteogenesis of the composite of the concentrated autologus bone marrow mesenchymal stem cells(MSCs) and find a potential method of treating the osteonecrosis of femoral head. Methods:Twenty - four Chinese white rabbits were divided randomly into three groups. The composite which consisted of the MSCs, BMP and DBCB was implanted into rabbit sacrospinalis muscle bags as designed. The MSCs concentration for group Ⅰ , Ⅱ and Ⅲ was 1 × 10^6, 1 × 10^5 and 1 ×10^4 respectively. The samples were harvested and examined radiographically, grossly and histologically at 2,4,8 and 12 weeks postoperatively. Results: The amount of the new bone was the largest in group Ⅰ. Conclusion: The amount of the new bone is positively related to the concentration of MSCs in the composite and its potential for treatment of ostconecrosis of femoral head remains to be studied.
