摘要
目的研究心肌营养素-1(cardiotrophin-1,CT-1)在血管紧张素II(AngⅡ)刺激心肌细胞造成肥大、损伤时的作用及意义。方法利用AngⅡ刺激心肌细胞造成细胞肥大,光、电镜下观察细胞形态变化,免疫组化法检测CT-1蛋白表达,逆转录聚合酶链反应(RT-PCR)检测CT-1、β-肌球蛋白重链(β-MHC)mRNA表达,酶联免疫法(ELISA)定量检测心肌细胞凋亡。结果经AngⅡ刺激后心肌细胞形态变大,β-MHCmRNA表达增加,心肌细胞凋亡增多;CT-1mRNA、蛋白表达均增加;苯亚甲基丙二腈的脂类衍生物(AG490)能部分抑制CT-1引起的心肌细胞肥大,并减少心肌细胞凋亡。结论CT-1参与心肌细胞肥大的过程;AG490能够抑制CT-1促心肌细胞肥大的作用,而不降低CT-1对心肌的保护作用。
Objective To investigate the effect and significance of cardiotrophin-1 (CT-1) in the hypertrophy and lesion of myocardial cells induced by angiotensin Ⅱ (Ang Ⅱ ), Methods The cultured myocardial cells were treated with Ang Ⅱ to induce hypertrophy. The morphologic changes of myocardial cells were observed under contrast phase microscope and electron microscope, The mRNA levels of CT-1 and β-MHC were examined by RT-PCR method, The protein expressions of CT-1 were examined by immunohistochemistry method, The apoptosis of myocardial cell was identified by ELISA, Results After stimulated by Ang Ⅱ , the myocardial cells enlarged, Both the mRNA levels of β-MHC and the apoptosis increased. The mRNA level and protein expression of CT-1 increased, AG490 may partially inhibited the myocardial cell hypertrophy and decreased the apoptosis related with CT-1 Conclusions CT-1 may take part in the hypertrophy of myocardial cells. AG490 inhibits the hypertrophy and does not decrease the protective effect of CT-1.
