摘要
目的观察中药清肠栓对实验性溃疡性结肠炎大鼠结肠上皮细胞凋亡率的影响,探讨其作用机制。方法用三硝基苯磺酸制备大鼠实验性溃疡性结肠炎模型,随机分为模型组、SASP组、清肠栓组、正常对照组,给药7d后处死,剪取远端结肠,流式细胞仪检测结肠上皮细胞凋亡率。结果正常组、SASP组与清肠栓组的结肠上皮细胞凋亡率均较模型组明显降低,差异均有非常显著性意义(P<0.01),清肠栓组凋亡率的平均值较SASP组凋亡率的平均值低,但两组凋亡率的差异没有统计学意义(P0.05)。结论中药清肠栓治疗溃疡性结肠炎的作用机理与降低结肠上皮细胞凋亡率有关。
Objective To investigate the influence of “Qingchang Suppository” on the apoptosis rate of colonic epithelia in rats with ulcerative colitis and its mechanism. Methods SD rats are injected with trinitro-benezen-ulfonicacid (TNBS) through anus to make ulcerative colitis. Model group, SASP group, normal group and herbal group were established and treated correspondingly. After seven days, the distal colon was isolated for apoptosis rate respectively with flow cytometry mechine. Results Apoptosis rate is significantly higher in model group than in normal group, SASP group and herbal group(P 〈 0.01 ) ; the average of apoptosis rate of herbal group is lower than that of SASP group, however the difference is not significant (P 〉 0.05). Conclusion It suggests that “Qingchang Suppository” acts to treat ulcerative colitis probably by decreasing the apoptosis rate of colonic epithelia.
