摘要
目的观察异氟醚麻醉后大鼠不同脑区、不同时期cAMP含量的动态变化,探讨cAMP在其麻醉中的作用。方法SD大鼠40只,分层随机设计均分成5组,每组8只,吸入1.5%异氟醚,分别在未吸入异氟醚时(对照组)、翻正反射即将消失时(诱导期组)、翻正反射消失后1 min(麻醉期组)、翻正反射恢复即刻(恢复期组)和翻正反射恢复后1 h(清醒期组)断头取脑。放射免疫法测定大脑皮层、海马和脑干的cAMP含量。结果在皮层,与对照组相比,cAMP含量在诱导期组即升高(P<0.05),麻醉期组进一步升高(P<0.01),恢复期组迅速下降至对照组水平。在海马,与对照组相比,各组cAMP含量均无明显变化。在脑干,与对照组比,仅麻醉期组cAMP含量升高(P<0.05),而其他组无显著变化。结论异氟醚升高大脑皮层及脑干cAMP含量(而对海马却无影响),提示cAMP在异氟醚的全麻机制中可能发挥重要作用。
Objective To investigate the effects of isoflurane on cyclic adenosine monophosphate (cAMP) content in cerebrocortex, hippocampus and brain stem of rats, and to examine the role of cAMP in the mechanism of isoflurane anesthesia. Methods Fourty SD rats were delaminatedly designed and allocated randomly to 5 groups inhaling 1.5 % isoflurane: no administration (in the control group, n = 8), rats' righting reflex was about to lose (the induction group, n = 8), after losing of righting reflex for one minitue (in the anesthesia group, n = 8), righting reflex was about to recover (in the recovery group, n = 8) and one hour after righting reflex recovery (aware group, n = 8). The brain tissues were rapidly dissected into cerebrocortex and hippocampus and brain stem. Then the cAMP content was determined, Results At cerebrocortex, compared with that in control group, the cAMP content in induction group were significantly increased (P 〈 0.05), and in anesthesia group continual increased (P 〈 0.01). At hippocampus, compared with that in the control group, there were no significant changes in the cAMP content in all groups. At brain stem, compared with that in the control group, the cAMP content only in anesthesia group was significantly increased (P 〈 0.05). Conclusion Isoflurane could increase the cAMP content in cerebrocortex and brain stem of rat (no effect on hippocampus), all the results indicate that cAMP may play an important role in the mechanism of isoflurane anesthesia.
