摘要
目的:观察丙泊酚麻醉后大鼠不同脑区、不同时期cAMP含量的动态变化,探讨cAMP在丙泊酚麻醉中的作用。方法:Sprague-Dawley(SD)大鼠40只,分层随机区组设计分成5组,每组8只,腹腔注射异丙酚100mg/kg。分别在腹腔注射生理盐水10ml/kg(对照组)、翻正反射即将消失时(诱导期组)、翻正反射消失后2min(麻醉期组)、翻正反射恢复即刻(恢复期组)和翻正反射恢复后1h(清醒期组)断头取脑。放射免疫法测定大脑皮质、海马和脑干的cAMP含量。结果:与对照组比较,诱导期组皮质和麻醉期组皮质、海马、脑干cAMP含量升高。与麻醉期组比较,恢复期组皮质和清醒期组皮质、海马、脑干cAMP含量降低(P<0.05或0.01)。结论:cAMP在丙泊酚的全麻机制中可能发挥重要作用。
Objective: To investigate the effects of propofol on cyclic adenosine monophosphate(cAMP) content in cerebrocortex, hippocampus and brain stem of rats,and to examine the role of cAMP in the mechanism of sevoflurane anesthesia.Methods: Fourty SD rats were delaminately designed and allocated randomly, to 5 groups paunch inject 100mg/kg propofoh paunch inject 0.9% NaCl 10mg/kg(control group, n = 8), rats' righting reflex was about to lose( induction groupA, n = 8; induction group B, n = 8), after losing of righting reflex for one minitue(anesthesia groupA, n = 8; anesthesia groupB, n = 8), righting reflex was about to recover(recovery groupA, n = 8; recovery groupB, n = 8) and one hour after righting reflex recovery (aware groupA, n = 8; aware groupB, n = 8). The brain tissues were rapidly dissected into eerebrocortex and hippocampus and brain stem. Then the cAMP content was assessed. Results: Compared with the control group, the cAMP content in the cerebrocortex, hippocampus and brain stem of induction and anesthesia group were increased; ompared with the anesthesia group, the cAMP content in the cerebrocortex, hippocampus and brain stem of recovery group and aware group were decreased( ( P 〈 0.05 orP 〈 0.01 ) ;Conclusion: cAMP may play a important role in the mechanism of sevoflurane anesthesia.
