摘要
目的研究移植肝细胞在放射性肝损伤(RILD)动物模型体内的代谢支持作用,评价肝细胞移植对RILD的疗效。方法用60Co照射实验大鼠肝区,总剂量40 Gy,一次性照射,剂量率为100 cGy/min,制成急性RILD动物模型。4 d后将所有照射动物施行部分肝叶切除术(约切除2/3)。以改良两步法分离同种属大鼠肝细胞,并立即将上述动物随机分为两组,实验组行经脾脏实质的肝细胞移植,移植细胞数为5.0×106;对照组经脾实质注射等量生理盐水。术后1个月两组分别采血测血清清蛋白水平。术后2个月再次检测清蛋白,并做常规病理切片及透射电镜检查,比较两组在组织结构上的差异。动态观察两组死亡率的变化。结果肝细胞移植术后1个月,实验组血清清蛋白水平明显上升,和对照组相比有显著差异。移植后2个月,实验组血清清蛋白水平进一步升高,与对照组、本组1个月前的水平均有显著差异。移植后1个月和2个月两组动物死亡率的差异均有统计学意义。组织学检查示实验组RILD轻于对照组。结论同种肝细胞经脾脏移植后,在RILD动物模型体内有良好的代谢支持作用,并可提高其短期生存率。
Objective To study the metabolic support action of transplanted hepatocytes in the model of radiation-induced liver damage(RILD), and to evaluate the therapeutic effects of transplanted hepatocytes in RILD. Methods The models were made by exposing rats liver to ^60Co irradiation, the dose was 40 Gy, and the dose ratio 100 cGy/min. Four days later, the rats underwent partial hepatectomy. Cells were isolated with a modified collagenase perfusion method. Animals were divided into two groups randomly. In the experimental group, 5.0 × 10^6 hepatocytes were injected into the spleens while the control underwent nature saline injection in the same places. Blood samples were obtained at an interval of one month to measure the serum albumin in the two groups, and histopathological changes were evaluated by pathological and electron microscopic examination. The survival rates in the two group were observed dynamically. Results One month after hepatocytes transplantation(HT), serum albumin level of the experimental group ascended obviously, having significant differences with that in the controls. Still one month later, it ascended progressively, showing significant differences with that in the controls as well as with itself 1 month ago. Both the differences of mortality rate in the two groups 1 month and 2 monthes after HT showed statistical significence. Histological examination suggested that radiative damage was ameliorated in the experimental group compared with that in the controls. Conclusion After transplanted into the spleen of the RILD model, congeneic rat hepatocyte can provide fairly good metabolic support and ameliorate the short-term survival rate in the model.
