HLA-G在胃癌组织中表达的研究分析 被引量:1

摘要 目的探讨HLA-G在肿瘤免疫逃逸中的作用。方法采用美国BD公司FACSCalibur检测仪及Cellquest分析软件,对经病理组织学确诊为胃癌患者的瘤组织、瘤旁正常组织、淋巴结转移组织进行HLA-G水平测定。结果在胃癌组织及转移性淋巴结中的表达较低,而在癌旁正常组织中呈高表达,胃癌组织与转移性淋巴结之间的表达无统计学意义(P>0.05)。癌旁正常组织、胃癌组织与转移性淋巴结两组间表达具有统计学意义(P<0.01)。结论癌旁正常组织中HLA-G表达高于癌组织,考虑癌组织可能通过HLA-G的低表达而逃逸机体免疫系统攻击;癌旁正常组织中的HLA-G的高表达可能是由癌组织刺激所致,可抑制癌旁正常组织的抗癌作用。在淋巴结转移组织中表达HLA-G的阳性细胞降低。提示有转移的淋巴结虽然存在着一定的细胞免疫应答水平,但免疫逃逸现象亦因肿瘤细胞对淋巴结的浸润而存在。 [Objective] To explore the effect of HLA-G in immune escape. [Methonds] HLA-G as related factors of immune escape, derived from gastric cancer patients were assayed in tumorous tissues, peri-tumorous normal tissues and the nodes of lymphatic metastasis with FACScallbur (Beeton Dickinson immunocytometry system, U.S.A), respectively. [Results] The alteration of HLA-G in tumorous tissues and nodes of lymphatic metastasis was declined than that in peri-tumorous normal tissues (P 〈0.01), however, between them there was no significant difference (P 〉0.05). [Coulusions] The expressions HLA-Geells in peri-tumorous normal tissues were higher than those in tumorous tissues, considering that the tumorous tissues might escape from the attack of immune system by the low expression of HLA-G. In peri-tumorous normal tissues, the expressions HLA-G are increased. It may be assumed that tumorous tissues can stimulate peri-tumorous normal tissues and lead to a possibility of escaping the attack of immune system. The expressions of HLA-G are lower, which indicate that nodes of lymphatic metastasis can exist the immune response in a way, but phenomenon of immune escape is existed as tumor cells soaked the nodes of lymph.
