摘要
本文提出了一种竞争法偶合反应伏安酶联免疫分析测定人血清乙型肝炎核心抗体(HBcAb)的新方法。该法将辣根过氧化物酶标记的乙型肝炎核心抗体(HBcAb-HRP)催化H_2O_2氧化部甲联苯胺(OT)的反应与邻甲联苯胺的氧化产物在电极上的还原反应相偶合,根据测定标记在HBcAb上的HRP的量,求得发生液相竞争免疫反应的HBcAb的含量。测定HBcAb-HRP及HBcAb的灵敏度均高于经典的酶联免疫吸附法(ELISA)。用所建立的方法对病人血清样品进行了测定,并与现行的ELISA法进行对照,两种方法相关性很好。
Anew voltammetric enzyme immunoas-say with coupled reaction for the determina-tion of HBcAb in human serum is developed. The method is based on coupling the oxidationof OT-H_2O_2 catalyzed by HBcAB-HRP Withthe electro-reduction of the oxidizing productof OT to measure the activity of HBcAb-HRP,and the latter reaction exhibits a sensi-tive second order derivative linear sweepvoltammetric wave at-0.58V(SCE)in BRbuffer, so HBcAb can be measured.Thesensi-tivity of the present method to detect HBcAb-HRP and HBcAb is higher than that byELISA absorption method,Serum samplesfrom some patients have been analyzed withsatisfactory results.
