摘要
为阐明 Alzheimer 病(AD)与载脂蛋白E(apoE)ε_4等位基因之间是否存在关联,对54例AD 患者(23例早发性 AD,31例迟发性 AD)、37例多发梗塞性痴呆(MID)患者、42例精神分裂症患者及147名正常人(<45岁者72名,>60岁者75名)的 apoE 基因进行分型,计算基因频率,并对疾病与 apoE 基因多态性进行了关联分析。结果显示,apoEε_4基因与早发性和迟发性 AD 均有显著关联,而且随ε_4基因剂量增加,AD 的患病率上升。MID、精神分裂症及正常人在 apoE 基因分布方面无显著性差异。提示 apoE ε_4基因具有影响 AD 发病的特异性。
54 cases with Alzheimer's disease (AD;early-onset AD,23;late-onset AD,31),37 cases with multi-infarctive dementia (MID),42 cases with schizophrenia and 147 normal controls (ages<45 years,72;ages>60 years,75) were collected to count the gene frequency of the apolipopro- tein E(apoE)alleles,and the association between AD and apoE gene polymorphism was analysed.The gene frequencies of the three apoE alleles detected were as follows:AD,ε_2=0.0278,ε_3=0.7222, ε_4=0.2500;MID,ε_2=0.1351,ε_3=0.7568,ε_4=0.1081;schizophrenia,ε_2=0.0476,ε_3=0.8690, ε_4=0.0835;normal controls,ε_2=0.0867,ε_3=0.8067,ε_4=0.1067.The association analysis suggested that the association with apoE ε_4 happened not only in late-onset AD,but also in early-onset AD.Fur- thermore,with the increment of ε_4 gne dose,the susceptibility to AD increased.There were no signifi- cant differences among MID,schizophrenia and normal controls.It suggested that the influence of apoE allele ε_4 on AD might be exclusive.
