摘要 目的:采用高效液相色谱法测定阿卡波糖发酵液中葡萄糖的含量。方法:用NH2柱(250mm×4.6mm,5u),以乙腈、磷酸盐缓冲液(70∶30)为流动相,流速1.5mL·min-1,柱温40℃,用示差检测器检测。结果:线性范围为0.5~1.5mg·mL-1(r=0.9997)。平均回收率为91.44%,RSD=1.16%(n=3)。结论:本法具有操作简便,分析快速准确等优点。 Objective:A HPLC method was established for the determination of glucose in Acarbose fermentation Liquid. glucose in Acarbose fermentation liquid can be separated on NH2 column with a mixture of acetonitrile, phosphate buffer (70:30) as mobile phase and with RID detector. The flow rate was 1.5 mL/min,Column temperature was 40℃. Results: The standard curve was linear in the concentration range 0.5-1.5mg/mL and the correlation coefficient was 0.9996. The average recovery was 91.44 % and 1.16%respectively. Conclusion: The method is simple,rapid and accurate.
