The influence of different pH and salt concentration on the extraction of anti-D antibody by column chromatography

摘要 目的探讨不同pH值和盐浓度对离子交换色谱从低效价血浆中提取抗-D抗体的影响及工艺。方法抗-D效价为1∶128的健康人血浆,经DEAE-SephadexA-50离子交换色谱柱,用不同pH值和浓度的磷酸盐缓冲液(PBS)洗脱,收集未吸附组分,检测相关指标。结果pH7.5组的抗-D效价和IgG回收率较低,但γ-球蛋白纯度较高;而pH5.8、0.05mol/LPBS组的抗-D效价和IgG回收率最高,单体和二聚体含量也高,但γ-球蛋白纯度相对较低。结论用pH5.8、0.05mol/LPBS洗脱,较适合从低效价血浆中提取抗-D抗体。 Purpose To investigate the influence of different pH and salt concentration on the extraction of anti-D antibody from the plasma containing anti-D at lower titer by column ion-exchange chromatography (CIEC). Methods The plasma containing 1:128 titer of anti-D was processed on CIEC with different pH and concentration of PB eluent. The preparation containing anti-D eluted from DEAE-Sephadex A-50 column was collected and analyzed. Results The IgG content and anti-D titer were lower in the pH 7.5 groups than those in the pH 5.8 groups, but the purity of y-globulin was relatively higher. The anti-D titer and IgG content of the preparation from pH 5.8, 0.05 mol/L PBS group were the highest and the percentage of IgG monomer and dimmer was also high. Conclusion CIEC method with pH 5.8, 0.05 mol/L PBS as eluent seems ideally suitable for the production of anti-D from the plasma containing this antibody at low titer.
