摘要
目的研究从低效价血浆中分离抗-D抗体的方法。方法用遗传表现型CCDee的RhD阳性O型红细胞制备血影细胞,经亲和层析法从抗-D含量为0.814μg/mL的健康人血浆中分离抗-D抗体,检测相关指标。结果经此法成功获得抗-D抗体,抗-D抗体回收率达到84.65%,所得制剂经检测激肽释放酶原激活剂含量、IgG单体加二聚体含量、抗补体活性及γ-球蛋白纯度等指标符合中国生物制品规程的要求。结论血影细胞亲和层析法简化了抗-D抗体提取和纯化步骤,提高了从低效价血浆中提纯抗-D抗体的回收率。
Objective To investigate the method of separating human anti-D antibody from its low titer plasma. Methods The ghost cells were prepared with the group O RhD positive red blood cell (genotype CCDee). Then anti-D antibody was sepaiated from the plasma iv which the content of anti-D was 0.814 μg/mL by the affinity chromatography (AC) with the ghost cells. Results The anti-D antibody was collected by this method and the recovery of anti-D was reached 85.87%. The content of prekallikrein activator (PKA), anticomplement activity (ACA), monomer/dimmer, and purity of γ-globulin were all reached the requirements of Chinese national standards for biologicals. Conclusion This method simplifies the process of sepaiation and purification of anti-D, and increases the recovery of anti-D separation.
