摘要
考察超氧化物歧化酶(SOD)口腔贴膜制备过程中温度和保温时间对SOD活性的影响及不同厚度药膜药物释放度的差异测定药膜中SOD的活性及其回收率、体外溶出、皮肤刺激性及药膜中SOD稳定性;并进行药膜重量差异检查和卫生学检查;建立制备动物口腔粘膜溃疡模型,从溃面直径和愈合期两个方面考察药物的疗效。结果表明65°C下保温2h,SOD基本保持原有的活性;药膜厚度增加释药时间会延长;Cu,Zn-SOD和EC-SOD药膜的药物含量分别为(504.69±27.31)U/cm2和(489.0±22.4)U/cm2,并在15min内药物全部溶出;无皮肤刺激性,4°C下保存一个月药膜中药物含量稳定;与空白组比较,SOD药膜能显著减小溃面直径,缩短溃疡愈合期。
To prepare SOD oral pellicle with PVA as main material, the influence of temperature and time on the activity of SOD and the release rate of different thickness of the pellicle were studied. SOD content, its recovery rate, skin stimulation, stability, weight difference and hygiene in the pellicle were investigated. Animal model of oral ulcer was constructed to determine the curative effect of the pellicle with the diameter of ulcer area and healing time. Results indicate that, SOD retained almost all its activity at 65 ℃ for 2 h, With the increase of pellice thickness, the drug release time was extended. The content of Cu, Zn-SOD and EC-SOD in the pellicle was (504.69±27.31) U/cm^2 and (489.0±22.4) U/cm^2 respectively. Within 15 mins SOD can be released completely. No skin stimulating was observed, and the activity of SOD in the pellicles was stable at 4 ℃ for 1 months. Compared with the control healing SOD pellicle can reduce the diameter of ulcer area, and shorten the healing period significantly. SOD pellicle studied is claimed to comply with the requirement of pharmaceutical usage, and had a notable curative effect on oral ulcer.
