摘要
枯草芽孢杆菌(Bacillus subtilis)LL18-4菌株分泌物中的类细菌素通过硫酸铵盐析法粗提后,经过Sephadex G-75、DEAE-Sephadex A 50和Sephacryl S-200HR分步柱层析,SDS-PAGE电泳检测到单一蛋白质条带,分子量约为60 kD.该类细菌素在酸性条件下及80%饱和度的硫酸铵溶液中具有较高的稳定性,可4℃放置3个月仍保持80%以上活性;对有机溶剂有一定的耐受性;Mg^(2+)能提高其抑菌活性.指数增长前期添加柑桔绿霉(Peniicilbumdiigitatum)病原真菌的培养液可以提高该类细菌素的分泌量.
A bacteriocin-like substance(BLS) produced by Bacillus subtilis LL18-4 was purified via ammonnium sulphate precipitation,Sephadex G-75,DEAE-Sephadex A 50 and Sepharyl S-200 HR chromatography, and in the final purification step,one active fraction was obtained and the molecular weight of the purified BLS determined by SDS-PAGE was about 60 kD.The BLS was able to resist the denaturation action of several organic solvents and the antifungal activity was increased when incubated with Mg^(2+).The fermentation ability of LL18-4 was improved when the fermentation of Peniicilbum diigitaturn was added in the early exponential growth phase.
