摘要
目前,国内尚无将体外转译体系应用于IL 2研究的报道。本文以长春1号小麦种制备麦胚无细胞体系,经与兔网织红细胞、腹水癌细胞两体系比较筛选,结果证明麦胚无细胞转译体系适于实验室应用。通过对ML A 144mRNA作转译活性鉴定,证明该系统可以合成具有生物活性的、完整的IL2分子。
Compared with the other two translation systems(rabbit reticulocyte lysate and Ehrlich ascite tumor lysate) in vitro, the wheat germ system was available for IL-2 mRNA translation. We had selected the translation conditions for maximal IL-2 production. in which IL-2 activity was found to be depended on the amount of MLA-144 RNA (0~120μg) or poly (A)^+ RNA (0~5μg) added in the 200μl translating solution. The maximal index of incorporation of protein synthesis was 10.99 and the maximal translation efficiency of IL-2 mRNA was two percentage compared with normal MLA-144 cells.
