mIL-12基因对结肠癌细胞凋亡的诱导作用 被引量:1

摘要 目的探讨白细胞介素12(IL12)基因的抗结肠癌作用机制。方法鼠结肠癌动物模型建立后,待瘤体达近0.5cm时,每只瘤内注射携mIL12基因逆转录病毒载体的包装细胞(PA317mIL12)1×107,于治疗后6h处死并取肿瘤组织行透射电镜、原位末端标记法(TUNEL)及流式细胞仪(FCM)检测。FCM除分析肿瘤DNA含量、观察凋亡峰外,还同时进行AnnexinⅤ检测。结果PA317mIL12瘤内注射后6h,电镜下即可见典型的“凋亡小体”。TUNEL检测也同样显示有呈棕色染色的凋亡阳性细胞出现,癌细胞凋亡指数达37.6%。FCM检测不仅显示了清晰的凋亡峰,且AnnexinⅤ检测也为阳性。结论PA317mIL12介导结肠癌细胞(C26)凋亡可能是IL12基因抗肿瘤作用的重要机制之一。 Objective To explore the anti-tumor action mechanism of interleukin(IL)-12 gene in colon adenocarcinoma. Methods The tumor model was established in mice by inoculate tumor tissue subcutaneously. When diameter of tumor reached about 0.5 cm, 1×10^7 PA317-mIL-12 cells were injected into each tumor. 6 h later, tumor was resected and cell apoptosis was examined by transmission electron microscope (TEM), TUNNEL and FCM (including tumor cell DNA content and Annexin V examination). Results The fresh tumor tissue that had been treated by PA317-mIL-12 for 6 h was examined with FCM, TEM and TUNEL. The results showed that apoptosis cell dyed with brown and its chromatin gathered in peripheral area of nuclear had presented, an obvious sub-Cn apoptosis peak had formed and Annexin V was positive. Apoptosis index was 37. 6%. Conclusion The cell apoptosis of colon cancer, mediated by PA317-mIL-12 may be one of the important antitumor mechanisms.
