消耗白细胞在兔油酸性肺损伤模型形成中的意义

Studies on the Role of Leukocyte-Depletion in the Formation of Oleic-Acid Induced Experimental Pulmonary Injury in Rabbits

摘要 用盐酸氮芥消耗兔白细胞90%左右,观察白细胞在兔油酸性肺水肿发病中的作用。实验分为正常组(5只),油酸组(5只),消耗白细胞组(5只)。与正常组比较,油酸组肺血流量在10min后各时间点均明显下降(P<0.05);而消耗白细胞组除30min点有一过性降低外(P<0.05),其它各时间点的变化均无统计学意义。肺湿干重比及肺水含量,正常组分别为4.91±0.20和79.56±0.79%;油酸组分别为5.95±0.4g(P<0.01)和83.12±1.46%(P<0.01);消耗白细胞组分别为6.77±1.32(P<0.05)和84.77±3.04%(P<0.01)。结果提示,白细胞是使兔油酸性肺水肿时肺血流量下降的原因之一,而对肺水肿程度影响不大。 Three groups of rabbits were usec: group I(n= 5), normal control;group Ⅱ ( n = 5 ) with oleic acid injection( iv ) ; group Ⅲ(n=5)with a single intravenous injection of nitrogen mustard (1.5mg/kg), 72-84 hours later, the leukocyte counting in the circulating blood of rabbits were decreased to about 10% of the original. Then oleic acid was injected into those rabbits. The pulmonary blood flow of each group was recorded directly with an electromagnetic blood flowmeter for 6 hours. Then all the rabbits were killed, the wet/dry lung weight ratio and lung water were calculated. As compared with groupⅠ , the pulmonary blood flow of group Ⅱ declined significantly( P<0.05)at any point of measurment 10 min utes after injection of oleic acid. At point of 30 minutes, the pulmonary blood flow of group Ⅲ was also less than that of group Ⅰ (P<0.05);but other points did not show significant differences. As to wet/dry lung weight ratio and lung water, group Ⅰ got 4.91±0.20, 79.56±0.79%; group Ⅱ 5.95±0.49(P<0.01), 83.12±1.46% (P<0.01); and groupⅢ 6.77±1.32 ( P<0 .05 ) , 84.77±3.04% (P<0.01) .These results suggest that the leukocytes may play a role in declining pulmonary blood flow in experimental oleic acid injury of rabbits, but they do not influence the degree of pulmonary edema.
