摘要
大赖草的成熟胚在含有2mg/L2,4-D的MS培养基上形成颗粒状愈伤组织。减少培养基中的2,4-D浓度,愈伤组织分化出绿色的芽点。生根培养基为不含任何激素的MS培养基。有分化能力的愈伤组织可以在保持培养基(MS+2mg/L2,4-D+0.5mg/LBAP)上长期保存。利用这一培养程序可以快速,大量地繁殖大赖草。文章还对外源激素在植物组织培养中的作用进行了讨论。
Mature embryos of Elymus giganteus formed nodulous calli on the MS medium containing2mg/L2,4-D. The calli started to differentiate green sports of buds after reducing the level of2,4-D in the medium. The medium for inducing roots of plantlets is the free MS medium. Thecalli which have the potentiality of differentiation can be maintained in the medium with 2 mg/L2,4-D and 0.5mg/L BAP for a long period. This cultured system can be used in the fast re-production in vitro of E. giganteus.The role which exagenous hormones play in plant tissue cul-ture was also discussed in this paper.
