摘要
以8种基因型的羊草幼穗为材料,研究了与愈伤组织诱导、状态调控、分化等过程相关的因素。结果表明,羊草愈伤组织的诱导率在不同基因型羊草幼穗之间存在较大差异,使用N6-MS培养基交替继代,培养基中添加2.0mg/L2,4-D、1.0mg/LABA、100mg/L水解酪蛋白(CH)、300~500mg/L谷氨酰胺(Gln)和500mg/L脯氨酸(Pro),可有效改善羊草愈伤组织的生长状态,使其保持良好的胚性并提高分化率。
With eight genotypes of Leymus Chinensis as the subject,induction, regulation and differentiation of callus were investigated in this experiment. The results showed that the different genotypes of Leymus Chinensis differed significantly in the induction rate,the growth states of callus were improved effectively,embryogenic status was better and the differentiation rate was increased when using N6-MS medium by turns,with 2.0 mg/L 2,4-D and 1.0 mg/L ABA,and supplying 100 mg/L CH,300-500 mg/L Gln and 500 mg/L Pro in subculture.
