摘要
应用杂交瘤技术,以2种抗原分别免疫BALB/C小鼠。从3次细胞融合中,筛选出4株分泌高滴度、高特异性抗A1血型物质单克隆抗体的杂交瘤细胞系。经多种免疫化学试验证明:其中B523-H10、D243-H92株杂交瘤扩大培养上清液仅凝集A1红细胞,而不凝集A2、B、O型红血胞。它们结合部位的结构互补于:人工接上脂肪链的A-活性三糖。从而表明该2株单抗为首次获得的抗A1亚型红细胞单克隆抗体。不仅为研究A1亚型血本质提供了极好的材料,也为医学临床提供A1亚型血分型试剂准备了条件。
Using hybridoma technique,We prepared a group of byhridomas by different immunization procedures.They secreted high level and high specific monoclonal antibodies to A1blood substances.It was demonstrated that the tissue culture superatant of two of them(D234-H9 and B523-H10)could only agglutinate human A1-type erythrocytes specifically,but could not agglutinate A2,B and O type erythrocytes by various immunochemical assays.The complemental structure of their combining site was most complementary to the synthetic A blood group nonreducing trisaccharides glycosidically linked to -O(CH2)8 COOH.The results shaw that this is the first report of two monoclonal antibodies to human A1-type erythrocytes which will offer excellent material for the study of the nature of A,subgroup blood.They can also offer the grouping reagents for A1 subgroup blood in clinical medicine.
