摘要
应用ACUSYST-Jr^(TM)中空纤维系统,生产抗人A型红细胞单克隆抗体。一次连续培养164天,生产含单抗培养液 32644 ml,抗体血凝效价和其他各项标准均达到临床检测标准。在连续培养过程中还对细胞生长和代谢的有关参数与抗体分泌的关系进行了研究和分析,为中试生产工程细胞的生物产品,建立了技术基础。
Hollow fiber system Acusyst-Jr?(Endotronica) was used for large scale cultivation of hybridoma cells SRBC-B1 to produce anti-A monoclonal antibodies. Before cultivation, the cell line was characterized to determine the optimal setpoints for the processor control system. During a 164-day cultivation period run in Acusyst-Jr, the cumulative amount of harvest was 30,000 plus ml with HA titer ranging from 1:64-1:256. The product samples were all verified to be suitable for the use as routine blood typing reagent without further concentration. Over the course of continuous cell cultivation and antibody production, the relationship between the cell metabolic parameters and the level of antibody secretion was analysed.
