摘要
采用流式细胞仪及逆转录-多聚酶链式反应技术对所建立的K562/Adm细胞株生物学特性进一步研究。结果表明该耐药细胞对化疗药物的摄取能力明显降低,细胞跨膜糖蛋白P-170及编码该蛋白的mdr-1mRNA具有过度表达。细胞酶学研究发现耐药细胞G6PD及6PGD活性明显升高,尤以G6PD为著。该细胞为多药耐药性及其相关机制的研究提供一良好模型。
Using flow cytometry and
reverse transcription polymerase chain reaction(RT PCR), the
biological characteristics of K562 multidrug resistant to
adriamycine(K562/Ad)cell line were identified. The remarkable
findings were the reduced intracellular accumulation
ofdaunorubicine(DNR),the overexpression of p glycoprotein(P gp)as
well as the mdr 1mRNAwhich encodes P gp in K562/Adm cell line in
comparison to its parental sensitive K562 cell line.enxymic analysis
showed that G6PD and 6GPD, especially the former elevated
significi1ntly. Ourresults indicated that K562/Adm cell line might be
used as an appropriate model for the study ofmultidrug resistance and
its related mechanisms.
