摘要
目的 :研究三根制剂对多药耐药 (MDR)细胞株的逆转作用。方法 :应用 MTT比色法 ,观察三根制剂对 MDR细胞的逆转作用 ,运用流式细胞仪 (FCM) ,测定其对耐药细胞积聚和外排阿霉素 (ADR)的影响。结果 :三根制剂可部分恢复K5 6 2 / ADR和 K5 6 2 / VCR耐药细胞对 ADR的敏感性 ,而对 VCR抗药性的逆转作用不明显 ;可增加 K5 6 2 / ADR和 K5 6 2 /VCR耐药细胞内 ADR的积聚 ,并对外排 ADR有一定影响。结论 :三根制剂可部分逆转耐药细胞的抗药性。
Obsjective:To observe the reversal effect of Sangen Mixture(SGM) in multidrug resistant(MDR) cell lines K562/ADR and K562/VCR.Methods:We used MTT assay to observe the mixture′s reversal effect.Then,we determined the effect of SGM on intracellular adriamycin(ADR) accumulation and drug efflux by flowcytometry(FCM).Results:10mg/ml,20mg/ml of SGM can reverse the K562/ADR cell′s resistance to adriamycin to some degree and 20mg/ml SGM can also partly reverse the K562/VCR cell′s resistance to adriamycin.10mg/ml,20mg/ml concentration of SGM can enhance the intracellular adriamycin accumulation in K562/ADR and K562/VCR cell lines.And also has the effect on the efflux of intracellular adriamycin in MDR cell lines.Conclusion:SGM can partly reverse the resistance on MDR cell lines. [
