摘要
本研究采用大鼠黄体细胞体外孵育技术,观察了缩血管肽内皮素-1(ET-1)对黄体细胞孕酮分泌的影响及其与细胞内信使cAMP的相互关系。结果表明,10-6mol/L的ET-l可显著抑制黄体细胞的基础孕酮分泌和hCG诱导的孕酮分泌;外源性cAMP和腺苷酸环化酶激动剂Forskolin则可促进黄体细胞分泌孕酮;当cAMP或Fonskolin与ET-1共同作用时,前者可完全反转ET-1对黄体细胞的抑制作用,孕酮分泌恢复到对照组水平。提示El-1可能是一种卵巢局部调节因子,通过抑制黄体细胞内的腺苷酸环化酶,减少cAMP的生成和释放,进而抑制黄体细胞的孕酮分泌。
Objective :To determine the effect of endothelin-l on the progesterone( P)ecretion by rat luteal cells and its relation with cAMP. Design :In vitro experimental study. Setting:Laboratory in Shanghai lnstitute of Planned Parenthood Research. Methods:Preparation of luteal cells suspension from Sprague-Dawley rat ovaries and incubation in KR-BGA1 solution,Samples of incubation medium were collected and stored at-20℃for measurement of Pand cAMP with radioimmunoassay. Results:Endothelin-l at a concentration of l0-6mol/L significantly reduced the basal and hCG inducedP secretion about 20%and29%respectively (P<0. 0l ).xogenous cAMP significantly increased the p se-cretion to about 22%of the control group(P<0.05).When added to the endothelin-1 treated medium theP secretion returned to the control level. Forskolin stimulated the P secretion to 2.l times higher than thecontrol (P<0. 0l ),and when added to the endothelin-l culture system,the P secretion increased to a levelhigher than the control (P<0.01).The cAMP concentration in the luteal cell mediuM was significantly re-duced(P<0.05)when culltured with endothelin-l. Conelusion :The study suggested that endothelin-1 may be an intra- ovarian regulatory factor which in-hibits progesterone secretion from rat luteal cells by suppressing the adenyl cyclase activity and reducingcAMP accumulation .
