药用作物掌叶半夏组织培养及药物成份分析 被引量:7

摘要 药用作物掌叶半夏(Pinellia Pedatisecta Schott)的种子在附加2,4-D0.5—6.0mg/L+BA0.5 mg/L的MS或B_5培养基上均能诱导出大量愈伤组织,但2,4-D的浓度变化对愈伤组织的形成和生长无明显影响,且无分化现象.愈伤组织生长快,分散性好,适于悬浮培养.种子在附加NAA0.1—4.0mg/L+BAO.5mg/L的MS培养基上均能形成愈伤组织并分化出根和芽,随NAA浓度降低,分化率增高.掌叶半夏愈伤组织经分析会有生物碱、β-谷甾醇、及多种氨基酸等成份,氨基酸含量多比半夏生药粉的含量高. Large amount callus were induced trom seeds of pinellia pedatisecta Schott cul-tured on MS or B5 medium containing 2,4-D 0.5-6.0 mg/ L and BA 0.5 mg/ L. Difference in 2,4-D concentration did not influence the inducing efficiency and the growth rate of the callus. The callus grew rapidly, with good dispersibility for suspension culture, but they did not differen-tiate. Callus induced from seeds on MS medium containing NAA 0.1-4.0 mg/L and BA 0.5 mg / L differentiated well. The differentiation frequency increased as the NAA concentration de-creased. Analysis of the callus and crude drug powder of p. pedatisecta Schott indicated that they all contained alkaloid, β-sitosterol and various kinds of amino acids, that the amino acid contents in the callus were more than those in the crude drug powder.
