摘要
以猕猴桃10个种42个样品叶片为试验材料,采用改良CTAB法提取基因组DNA,优化确立RAPD-PCR的反应体系。从80个随机引物中筛选出38个引物进行扩增,应用SPSS11.0forWindows软件进行聚类分析。结果表明,美味猕猴桃和中华猕猴桃亲缘关系近,可将其归为同一个种;品种之间确有按原产地聚类的趋势;“海沃德”与其营养系品种“皖翠”有14个引物扩增出了18条特异性条带,变异条带较多。
The genomic DNA of 10 Actinidia species including 42 samples were extracted by the improved CTAB method. The parameters in RAPD-PCR were optimized. Thirty-eight arbitrary primers used in the study were screened from 80 ones. According to the data of RAPD-PCR,the cluster analysis was done by the SPSS (11.0) for Windows. The results showed that the relationship between A.chinensis and A.deliciosa was so close that they should be considered as the same species, and the classification between cultivars was accordant with their geographical distribution, and 18 characteristic bands between 'Hayward'and 'Wancui'were amplified in 14 primers by RAPD-PCR.
