肺癌组织中诱导型一氧化氮合酶和血管内皮生长因子的表达及其与血管生成的关系 被引量:7

摘要 目的:研究诱导型一氧化氮合酶(iNOS)和血管内皮生长因子(VEGF)在肺癌组织中的表达及其与间质血管生成的关系。方法:应用原位分子杂交和免疫组织化学方法检测41例肺癌组织中iNOS和VEGF的表达,图像分析技术进行定量分析;CD34标记微血管内皮细胞,显微镜下计数微血管密度(MVD)。结果:肺癌组织中iNOSmRNA及其蛋白表达阳性率分别为60.98%和58.54%,明显高于癌旁(均为20%)及正常肺组织(均为16.67%)(P<0.005),且与淋巴结转移及临床分期有关(P<0.05);iNOSmRNA及蛋白表达与VEGF呈明显正相关关系(r=0.4125,P<0.01;r=0.4623,P<0.005),且iNOS和VEGF均与MVD明显相关(P<0.05)。结论:iNOS表达可以作为评价肺癌分期、预测转移风险的参考指标;iNOS与VEGF在诱导血管生成过程中可能发挥相互依赖的正调控作用。 Objective To investigate inducible nitric-oxide synthase (iNOS) and vascular endothelial growth factor (VEGF) expressions in lung cancinoma tissues and their relationships with angiogenesis. Methods The expressions of iNOS mRNA and protein, VEGF and tumor microvascular density (MVD) were detected in 41 lung carcinoma, 10 peri-carcinoma and 6 normal tissues by the methods of in situ hybridization and immunohistochemistry. Meanwhile, their correlations with angiogenesis and tumor clinical parameters were systematically discussed and analyzed. Results The expressions of iNOS mRNA and protein in lung carcinoma tissues were significantly higher (60.98% and 58.54%,respectively)than those of peri-carcinoma tissues(20%) and normal tissues(16.67%)(P<0.005), which associated with lymph node metastasis and clinical stages (P<0.05). There was obviously positive correlation between iNOS mRNA, iNOS protein and VEGF (r=0.412 5,P<0.01; r=0.462 3, P<0.005). Both iNOS and VEGF are associated with MVD (P<0.05). Conclusion iNOS expression can be used to evaluate clinical stages and predict the risk of metastasis of lung carcinoma. iNOS and VEGF may exert dependable interactions in angiogenesis.
