摘要
目的观察环境内分泌干扰物壬基酚(nonylphenol,NP)对大鼠腺垂体细胞增殖的影响。方法大鼠腺垂体细胞在DMEM/F12培养液(含10%胎牛血清)中采用单层贴壁培养。实验设空白对照、雌激素(E2)对照及NP4个剂量组,采用MTT法、流式细胞术及免疫细胞化学对大鼠腺垂体细胞增殖情况进行分析。结果与空白对照组相比,1×10-6mol/LNP抑制腺垂体细胞增殖。随着NP作用剂量提高,其抑制腺垂体细胞的增殖,且呈现出剂量—反应关系(P<0.05)。结论高剂量NP影响大鼠腺垂体细胞正常增殖,腺垂体细胞是NP作用的靶细胞之一。
Objective To know the effect of endocrine disrupting chemicals,nonylphenol,on proliferation of rat anterior pituitary cells. Methods The rat anterior pituitary cells were grown in DMEM/F12 medium containing 10% FBS,NP group in four kind of concentration ,positive control group (E2) and negative control group were set,and the proliferation of rat anterior pituitary cells were analyzed by MTT assay, flow cytometry and immunocytochemistry. Results Compared with the negative control group, the proliferation of rat anterior pituitary cells treated with nonylphenol (1×10 -6 mol/L) was significantly inhibited with the adding of NP concentration, the result of its inhibiting proliferation showed dose-response relationship (P<0.05).Conclusions The high concentration of NP interferes the proliferation of rat anterior pituitary cells,and the findings give a support to the hypothesis that anterior pituitary cells is the target cell for NP.
