摘要
目的利用芯片技术探讨壬基酚(nonylphenol,NP)对F1代脑组织发育相关基因表达影响。方法NP染毒F0亲代母鼠,分别提取NP组和对照组F1代雄性2d龄大鼠的脑组织mRNA,分别用cy5和cy3逆转录标记,与BiostarR-40S基因表达谱芯片杂交,并对cy3、cy5荧光信号做扫描分析。结果芯片结果显示实验组已知功能差异表达基因有16条,其中下调基因有13条,5条与脑组织功能及发育密切相关。结论NP对发育中的脑组织可产生多方面的影响,主要干扰神经细胞的代谢、神经递质的合成释放以及神经元的发育分化。
Objective To explore the effects of nonylphenol on brain development-associated gene expression profiles of F1 generation rats by means of microarry technique. Methods The mRNA were extracted respectively from the brain of 2-day-old F1 generation rats whose F0 male generation were tested with and without nonylphenol, then reversely transcribed to cDNA and labeled with cy5 and cy3 fluorescence. Subsequently,the cDNA probes were hybridized to the Mouse40S cDNA microarray and the fluorescent signals of cy5 and cy3 were scaned and analyzed. Results A total of 16 identified genes expressed differently, including 13 down-regulated in which 5 were related to brain function and development. Conclusions Nonylphenol can effect the brain function in many ways, mainly disturb metabolism, development and differentiation of neurocyte, and synthesis and release of neurotransmitter.
