三种大鼠神经病理性疼痛模型的制备和效果比较 被引量:43

Comparison of the establishment and afficacy of three neuropathic pain models

摘要 目的比较慢性坐骨神经缩窄损伤(CCI)、脊神经结扎(SNL)、保留性神经损伤(SNI)三种神经病理性疼痛模型产生疼痛的效果.方法选择150~200 g的SD大鼠19只,随机分为四组(对照组、CCI组、SNL组、SNI组),对照组(n=4)切开左后肢皮肤和肌肉,暴露坐骨神经后立即缝合,CCI组(n=4)、SNL组(n=4)和SNI组(n=7)分别于左后肢制作CCI、SNL、SNI疼痛模型.术前3 d至术后11 d隔日使用机械和热辐射刺激测定术侧后趾,比较它们对疼痛刺激的反应差异.结果CCI、SNL、SNI疼痛模型50%缩爪阈值于手术7 d后趋于平稳,各组内相比差异无显著性(P>0.05).术后7 d对照组、CCI组、SNL组、SNI组50%缩爪阈值分别为(14.13土1.49)、(2.55±0.47)、(1.52±0.58)和(0.79±0.36)g,大小依次为对照组>CCI组>SNL组>SNI组,差异有显著性(P<0.05).CCI组对热辐射刺激的缩腿潜伏期的差异性评分低于对照组,差异有显著性(P<0.05);但SNL组和SNI组对热辐射刺激的缩腿潜伏期的差异性评分与对照组相比差异无显著性.CCI组、SNL组、SNI组对热辐射刺激所产生的抬腿时间均高于对照组,差异有显著性(P<0.05).结论SNI疼痛模型不仅制作简单,而且效果优于SNL、CCI模型,并仅对机械刺激敏感. Objective To compare the establishment and afficacy of three neuropathic pain models:chronic constriction injury of sciatic nerve(CCI), spinal nerve ligation (SNL) and spared nerve injury(SNI). Methods Nineteen female SD rats (weighted from 150 to 200 g ) were randomized into four groups(group C,group CCI, group SNL, group SNI). The sciatic nerve of the left hind leg was ~exposed without any injury in group C, then the incision was sutured immediately. Group CCI was prepared according to the method described by Gary,group SNL according to Sun, and group SNI according to Isabelle. During the development of allodynia and ~hyperalgesia, touch stimulator and radiant heat stimulator were used to measure the response of operative hind paw to mechanical and radiant stimulation. Results During experiment, any sign of ~mechanical or heat allodynia did not occur in group C. But the surgical procedure in group CCI, SNL and SNI produced an apparent and stable hyperalgesia to noxious heat and mechanical stimulation since the 7th day after operation. On the 7th day after operation, the 50% paw withdraw thresholds in group C,CCI, SNL and SNI were 14.13±1.49,2.55±0.47,1.52±0.58 and 0.79±0.36 respectively,with an quantity order of group C>CCI>SNL>SNI(P<0.05). The difference in scores responded more rapidly to radiant heat stimulation was lower in group CCI than that in group C. But there was not any difference between group SNI and group C in responce to radiant heat stimulation.Conclusion Among three pain models, SNI was not only model easiely established, but also the most sensitive to ~mechanical stimulation(but not radiant heat stimulation).
