精子蛋白17mRNA在卵巢癌中的表达及临床意义 被引量:11

摘要 目的:测定精子蛋白(Sp)17mRNA在肿瘤组织中表达情况。方法:设计合成特异性引物,建立测定人Sp17mRNA的RTPCR的方法,以β肌动蛋白(βactin)作为内标。检测13份原发卵巢癌组织标本、3份卵巢癌腹水标本和13份其他肿瘤组织标本。用正常睾丸组织RNA作模板进行RTPCR作为阳性对照,正常卵巢组织为阴性对照。阳性对照可出现2个条带,其中Sp17扩增产物近500bp,βactin产物234bp。阴性对照仅出现βactin条带。结果:13份原发卵巢癌组织标本中,11例表达Sp17mRNA,阳性率为84.6%。3份卵巢癌腹水标本中,1份检出Sp17mRNA。其他肿瘤组织标本全部阴性。结论:Sp17mRNA在原发卵巢恶性肿瘤中高水平表达,可以作为临床诊断的辅助指标。 Objective:To determine the distribution of sperm protein 17 mRNA in variant kinds of neoplasms. Methods:A pair of primers was synthesized and a RT-PCR method for human sperm protein 17 was established with β-actin as internal control. Then 13 tissue specimens of ovarian carcinoma, 3 ascites specimens of patients with ovarian carcinoma, 2 ovarian carcinoma cell lines, and 13 other tumor tissue samples were detected. Results: Normal testis RNA was used as template for RT-PCR, and we got a Sp17 amplification product of approximately 500 bp and a β-actin amplification product of 234 bp. Negative control was normal ovary RNA with β-actin fragment only. Sp17 mRNA was detectable in 11 ovarian carcinoma tissue samples (84.6%, 11/13). One (1/3) of ascites was positive for Sp17 mRNA, while all the other tissues including ovarian fibroma and lymphoma were negative. Conclusion: Sp17 mRNA is abundantly expressed in malignant ovarian tumor, so Sp17 is likely to be a diagnostic marker and a target of immunotherapy for ovarian carcinoma.
