低温对人外周血淋巴细胞的同步化作用 被引量:8

EFFECT OF LOW TEMPERATURE ON CELL SYNCHRONIZED DIVISION IN PERIPHERAL BLOOD LYMPHOCYTES

摘要 目的:探讨低温对人外周血淋巴细胞的同步化作用。方法:将人外周血淋巴细胞在37℃培养66h,入4℃冰箱4h,再分别入37℃培养箱恢复培养0min、10min、20min、30min、40min、50min、60min,常规制片。结果:不恢复培养(0min)或短时恢复培养(10min、20min)组分裂指数最高,早期染色体数目最多。结论:4℃低温可提高有丝分裂指数,增加早期染色体数目。 Objective:To observe the effect of low temperature on cell synchronized division in peripheral blood lymphocytes.Methods:Peripheral blood lymphocytes were cultivated at 37℃for 66h, 4℃for 4h, then resumed culture at 37℃ for 0min, 10 min, 20 min, 30 min, 40 min, 50 min and 60 min respectively. At last, the specimen were prepared as usual.Results:The highest mitotic index and the most number of early phase chromosomes appeared in 0 min, 10 min and 20 min resumed culture group. Conclusions: Low temperature treatment at 4℃ can increase the mitotic index and the number of early phase chromosomes.
