摘要
背景:研究发现多种蛋白的异常表达与脑胶质瘤的发生、发展密切相关,而CyclinD1蛋白的异常表达与其发生、发展及预后的关系尚不清楚,有待于进一步研究。目的:探讨脑胶质瘤组织中CyclinD1蛋白异常表达的变化,以及与肿瘤危害程度的关系。设计:以病理标本为研究对象的对照研究。单位:一所大学医院的神经外科。对象:48块不同恶性程度的脑胶质瘤标本来自1990-01/1995-12在西安交通大学第二医院神经外科住院手术治疗的脑胶质瘤患者,12块正常脑组织来自1990-01/1995-12在西安交通大学第二医院神经外科住院行内减压的非肿瘤患者。方法:用S-P免疫组化技术检测CyclinD1蛋白的异常表达,同时将染色结果与患者临床特点密切联系作以对比分析。结果:CyclinD1蛋白在人脑胶质瘤组织中的阳性表达率为54.12%,在正常脑组织中的阳性表达率为8.33%,二者差异有显著性意义(χ2=8.1481,P=0.0043)。在较低恶性程度人脑胶质瘤组织中的阳性表达率为37.04%,在较高恶性程度人脑胶质瘤组织中的阳性表达率为76.19%,二者差异有显著性意义(χ2=7.2940,P=0.0069)。在术后生存期较长患者的人脑胶质瘤组织中CyclinD1蛋白阳性表达率为70.37%,在术后生存期较短人脑胶质瘤组织中阳性表达率为33.33%,二者差异有显著性意义(χ2=6.5268,P=0.0106)。
BACKGROUND:Research has showed that the abnormal expression of some proteins c losely relates to the occurrence and development of cerebral glioma.However,the relationship between the abnormal expression of CyclinD1 protein and the occurre nce, development and prognosis of glioma is still uncertain which needs further study. OBJECTIVE:To study the expression of CyclinD1 protein in cerebral glioma and r elationship between it and the impact of tumor DESIGN:Control study based on pathological specimens. SETTING:Neurosurgery department of a university hospital. PARTICIPANTS:Totally 48 glioma specimens of different malignant degree were co llected from the patients who accepted surgery treatment in Neurosurgery Departm ent of Second Hospital of Xi'an Jiaotong University during January 1990 and Dece mber 1995.Twelve normal cerebral specimens were from the non tumor patients who were conducted intracranial pressure reducing in Neurosurgery Department of Sec ond Hospital of Xi'an Jiaotong University during January 1990 and December 1995. METHODS:S P immunohistochemical technique was used to detect the abnormal exp ression of Cyclin D1 protein. Simultaneously, the dyeing results and clinical ch aracters of patients were associated in order to conduct comparison. RESULTS:The positive expression rate of CyclinD1 protein in human cerebral gli oma was 54.12%while in normal cerebral tissue it was about 8.33%.There was sig nificant difference between them(χ2=8.148 1,P=0.004 3).And the positive express ion rate in cerebral glioma of low malignancy was 37.04%while in specimens of h igh malignancy it was 76.19%,there was significant difference (χ2=7.294 0, P=0 .006 9).The positive expression rate of CyclinD1 protein in specimens of patient s with long survival period and short survival period after surgery was 70.37%a nd 33.33%respectively with significant difference between them (χ2=6.526 8,P=0 .010 6). CONCLUSION: CyclinD1 is closely related to the occurrence and development of h uman cerebral glioma. It has provided experimental evidence for the prevention t o the occurrence of glioma and the estimation of its prognosis by studying the a bnormal proliferation of glioma cells targeted on CyclinD1.
