摘要
目的 :通过观察脑胶质瘤中 p16mRNA及其蛋白、EGFR的表达 ,探讨其在脑胶质瘤发生、发展中的作用。 方法 :应用原位杂交技术和免疫组织化学方法在 38例人脑胶质瘤中检测 p16mRNA及其蛋白、EGFR的表达。以 7例正常组织作为对照。结果 :p16mRNA和 p16蛋白在Ⅲ~Ⅳ级、Ⅰ~Ⅱ级胶质瘤、正常组织中的阳性表达率分别为 8 3% ,16 7% ;5 7 7% ,5 7 7% ;71 4% ,85 7%。EGFR在Ⅲ~Ⅳ级、Ⅰ~Ⅱ级胶质瘤、正常组织中的阳性表达率分别为 83 3% ,38 5 % ,14 3%。结论 :p16基因缺失引起的 p16蛋白的表达降低、EGFR的过表达在脑胶质瘤的发生中起重要的作用。
Purpose To investigate the effects of p16 mRNA and its protein, EGFR in the tumorigenesis of the cerebral glioma.Methods The expression of p16 mRNA and its protein, EGFR was detected in 38 cases of the cerebral glioma using in situ hybridization and immunohistochemical staining Seven normal brain tissues were used as control Results The positive expression rate of p16 mRNA and p16 protein in Ⅲ~Ⅳ、Ⅰ~Ⅱgroups of glioma and normal tissues was 8 3%, 16 7% ; 57 7%,57 7% ; 71 4%, 85 7%, respectively The positive expression rate of EGFR in Ⅲ~Ⅳ、Ⅰ~Ⅱgroups of glioma and normal tissues was 83 3%, 38 5%, 14 3%, respectively Conclusion Descended expression of p16 protein is due to the deletion of p16 mRNA Descended expression of p16 protein and excessive expression of EGFR may play an important role in the tumorigenesis of cerebral glioma
