摘要
目的 观察环氧合酶2(COX 2)的特异性抑制剂NS 398对乳腺癌MCF 7细胞的生长抑制和凋亡作用,及其对阿霉素(ADM)诱导的MCF 7细胞凋亡作用的影响。方法 应用四甲基偶氮唑蓝(MTT)比色法分析NS 398 对MCF 7 细胞的生长抑制作用,进一步用NS 398和ADM单独及联合作用于MCF 7细胞,流式细胞术检测细胞的凋亡,免疫印迹法检测凋亡相关蛋白Bcl 2、Bax的表达。结果 40μg/ml的NS 398就可抑制MCF 7 细胞生长、诱导其凋亡,下调Bcl 2 蛋白表达;和ADM联用凋亡率显著增高。结论 NS 398可抑制MCF 7细胞生长、诱导其凋亡,并且和阿霉素有协同作用,其诱导凋亡与Bcl 2表达下调有关。
Objective To investigate the effect of COX-2's selective inhibitor, NS-398,on growth and apoptosis induction of breast cancer cell line(MCF-7).Also,we examined whether NS-398 augments the sensitivity of MCF-7 to adriamycin.Methods Estrogen receptor(ER)-positive MCF-7 human breast cancer cell line was treated by NS-398 or adriamycin alone,or in combination.Cell proliferation was evaluated by MTT assay.Rate of apoptosis was determined by flow cytometry.Expression of Bcl-2 and bax was evaluated by western blotting.Results Incubation with NS-398 40 μg/ml ,MCF-7 cells exhibited significant proliferation inhibition,induction of it's apoptosis,and the expression of Bcl-2 was markedly reduced.When NS-398 combined with adriamycin,the rate of apoptosis was surprisingly enhanced.Conclusion NS-398 can inhibit MCF-7 cells proliferation and induce i'ts apoptosis,and there is synergism between NS-398 and adriamycin.Induction of apoptosis was associated with Bcl-2.
