摘要
目的 研究槲皮素对人肺腺癌A549细胞增殖和凋亡的影响。方法 采用MTT试验、流式细胞仪检测槲皮素对体外培养的人肺腺癌A549细胞的抑制作用和细胞周期的变化,电镜观察超微结构变化,免疫组化技术检测槲皮素对凋亡相关蛋白Bcl 2、p53表达的影响。结果 MTT结果显示槲皮素对体外培养的A549 具有明显的增殖抑制作用,且具有一定的浓度和时间依赖性;流式细胞仪检测发现,30μg /ml 槲皮素作用A549细胞48h后能将细胞阻滞于G0/G1 期, 且在细胞周期G1 峰的左侧出现了凋亡峰;电镜观察发现有凋亡的特征性改变;免疫组化结果显示,槲皮素使Bcl 2 表达下调,而上调p53 蛋白的表达。结论 槲皮素能抑制肺腺癌A549细胞的生长,阻止细胞由G0/G1 期向S期和G2/M期移行并诱发细胞凋亡,其诱发A549 细胞凋亡的机制可能与上调促凋亡蛋白p53表达和下调凋亡抑制蛋白Bcl 2的表达有关。
Objective To study quercetin′s effect on proliferation and apoptosis of A549 lung cancer cell.Methods The inhibition of quercetin on A549 cell was evaluated by means of MTT, cell cycle and apoptosis were observed by flow cytometry and electron microscopy, the expression of P53 and Bcl-2 were analyzed by immunocytochemistry.Results Quercetin inhibited cell growth in concentration and time dependent manners, prevented A549 cell entering G2/M+S from G0/G1 phase, and induced apoptosis. In the result of immunocytochemistry quercetin induced up-regulation expression of P53 and down-regulation expression of Bcl-2.Conclusion Quercetin can inhibit growth and induce apoptosis of lung cancer A549 cell, cell cycle is controlled in G0/G1. Meanwhile, quercetin can induce up-regulation expression of p53 and down-regulation expression of Bcl-2,which may be the mechanism of apoptosis of lung cancer cells by quercetin.
