摘要
从11份土样中分离出109株具碱性蛋白酶活力菌株。从中筛选出一株枯草芽孢杆菌(BacillusSubtilis)109号具高温碱性蛋白酶(TAP)活力,在pH10环境中好氧生长,最适生长温度为37℃左右,TAP活力平均为86.9U/mL。109号菌株经亚硝基胍诱变后,得到一株TAP活力提高至301U/mL的突变株M92,该诱变株再用利福平抗性平皿自然分离得到一株具利福平抗性菌株R01,其TAP活力达350U/mL。
strain of bacteria with the activity of TAP were isolated. A mutant strainR01 with high yield of TAP and stability was obtained from wild Bacillus subtilis 109 bytreatment of 78 mg/L NTG and 0. 3  ̄ 0. 4 mg/L Rifampen. The enzyme activity Increased from 86. 9 U/mL to 350. 2 U/ml- after heat treatment at 70'C for 30 min.
