摘要
嗜碱性的短小芽孢杆菌 R115(Alkaliphillc Bacillus pumilus)经0.4mg/ml亚硝基胍和0.4μg/ml 利福平处理,获得一株具有高产稳产碱性蛋白酶的变异株(B45),产酶活力由2803u/ml 提高到6000u/ml(28℃测定),该诱变株的最适产酶条件为起始 pH10.5—11.0,温度30—35℃,培养时间52—54h。0.4—0.6%K_2HPO_4可增加酶的产量。
A mutant strain B45 with high yield ofalkaline proteinase and stability was obtainedfrom alkaliphilic Bacillus pumilus R115 bytreatment of 0.4 mg/ml MNNG and 0.4 μg/ml Rifampicin.The enzyme activity was im-proved from 2803 u/ml to 6000 u/ml (assayedat 28℃).The optimum conditions for enzymeproduction were:initial pH 10.5—11.0,30℃for 52—54 h.0.4--0.6% K_2HPO_4 had a posi-tive effect on the enzyme production.
