酶解压片分离沙田柚胚襄与胚胎发育研究 被引量:4

摘要 用2.5%纤维素酶与2.5%果胶酶混合液在30℃下对已固定的沙田柚(Citrusgrandisvar.shatinyuHort.)胚乳酶解6—10小时,分离了开花前、授粉至受精间隔期、合子休眠期、胚和胚乳发育各期的胶囊.用Olympas相差显微装置进行观察与摄影,在胚囊中显示了沙田柚胚胎发育各主要阶段的结构特征。 The ovules from fixed Shatinyu (Citrus grandis var. shotinyu Hort.)were enzymicly macerated in a mixed solution of 2.5% pectinase and 2.5% cellulase at 30℃ for 6-10 hours. The embryo sacs in the stages before blooming, between pollination and fertilization,and of the zygote dormancy, and in various developmental stages of embryo and endosperm, were isolated. By means of OLYMPAS phase contrast microscope, the embryo sacs were observed and photographs were taken.The structural characteristics of every major stages of the embryonic development were shown in the embryo sacs. The prospects of the enzyme. squash technique for studying the embryonic development of citrus were analysed.
