摘要
目的:为进一步研究运动训练过程中心肌线粒体功能的变化.方法:观察不同时间跑台运动对大鼠心肌线粒体H+转运ATP酶活性及膜流动性的影响.结果:大鼠在运动至15min时,心肌线粒体H+转运ATP酶活性及寡霉素的抑制率与对照组比较无明显变化,线粒体膜的荧光偏振度明显降低(0.279±0.008vs0.267±0.007,P<0.05);大鼠在运动至衰竭时,心肌线粒体H+转运ATP酶活性及寡霉素的抑制率与对照组比较均明显降低[6.86±1.64vs4.56±0.46μmol/(min·mgpr)和(94.29±3.74)%vs(48.19±14.33)%,P<0.01],线粒体膜荧光偏振度降低显著(0.279±0.008和0.267±0.007vs0.258±0.010,P<0.01).结论:一定强度下不同时间的运动训练,不足以使心肌线粒体功能发生明显改变,但一次力竭性运动可以导致心肌线粒体膜的流动性增强,H+转运ATP酶活性降低.
Objective:To study the changes of mitochondrial function in myocardiocytes during exercise. Methods:Twenty-seven male SD rats were randomized into 3 groups. Group 1 served as the control group,and rats of Group 2 and 3 ran on treadmill at a speed of 27 m/min and an angle of 8 for 15 min and for as long as physical exhaustion occurred,respectively. Immediately after exercise , hearts were collected for examina-tion of the fluidity of submitochondrial membrane in myocardiocytes,the activity of H+ -transporting ATPase and its sensitivity to oligomycin,Results:When rats ran up to 15 min,the fluidity of submitochondrial mem-brane of myocardiocytes was significantly greater than that of the control(0.279±0.008 vs 0.267±0.007,P<0.05),and there was no significant difference between Group 2 and the control in the activity of H+ -trans-porting ATPase in mitochondria of myocardiocytes and its sensitivity to oligomycin.When rats ran to physi-cal exhaustion,there was a significant reduction in the activity of H+ -transporting ATPase in mitochondria of myocardiocytes and its sensitivity to oligomycin[6.86± 1.64 vs 4.56±0.46μmol/(min·mg protein)and 94.29±3.74 vs 48.19±14.33%, P<0.01],and the increase in membrane fluidity was greater than that of Group 1 and Group 2(0.279±0.008 and 0.267±0.007 vs 0. 258±0.010,P<0.01),respectively. Conclu-sions:The aforementioned findings indicated that one bout of exhaustive exercise might cause changes in the fluidity of mitochondrial membrane in myocardiocytes and a significant reduction in the activity of H+ -trans-porting ATPase while physical training of some intensity was not adequate in inducing apparent changes in the function of mitochondria in myocardiocyte.
