摘要
本文报道测定16种抗卫氏并殖吸虫(Pw)囊蚴和成虫单克隆抗体(McAb)的同型及其交叉反应。其中,抗Pw成虫McAb A_3D_3为IgM,k型。应用Dot-ELISA方法,以McAb A_3D_3检测痰卵阳性患者21例和痰卵阴性疑似患者63例血清循环抗原(CAg)),结果痰卵阳性患者CAg阳性率为100%(21/21),痰卵阴性疑似患者CAg阳性率为85.7%(54/63);同法检测45例日本血吸虫病、55例华支睾吸虫病、24例丝虫病、49例肠线虫感染、31例肺结核患者以及93例健康人对照血清均为阴性。结果表明,McAb A_3D_3的特异性及敏感性强,因而在Pw活动性感染和Pw病的早期诊断上均具有应用和推广前景。
The present paper reports on the screening sixteen monoclonal antibodies against either metacercaria or adult worm of Paragonimus westermani (Pw) with immunoglobulin classi- fication and further cross-reaction detection. A monoclonal antibody (McAb),A_3, D_3,to soluble antigen of Pw adult worms is IgM,type k. It has strong positive reaction with metabolic antigens of both Pw adult and adolecent worms;it has not any significant cross-reaction with antigens of Pw metacercariae, Pa. skrjabini,P. heterotremus,Schistosoma japonicum,Clonorchis sinensis, Fasciolopsis buski,Fasciola hepatica, Ascaris Lumbricoides adult worm, S. mansoni and japonicum SEA, Brugia malayi microfilariae,leishmania major promastigotes and phosphorylcholine conju- gated bovine serum albumin (PC-BSA). Pw circulatings antigen in sera from 75 patients with positive (21 cases) or negative (54 cas- es)egg detection in sputa have been successfully detected with A_3D_3 Dot-ELISA. No Pw circulat- ing antigen was detected in sera from 173 other parasitosis (45 schistosomiasis japonica, 55 clonorchiasis sinensis, 24 filariasis, 49 intestinal helminthiasis) , 31 pulmonary tuberculosis cases, and 93 normal individuals. Positive detection rates in 21 patients with positive egg detection and 63 suspicious patients with negative egg detection were 100%(21/21) and 85. 7%(54/63) respec- tively. The results indicated that A_3D_3, has high specificity and sensitivity, it has strong reaction whth metabolic antigen of Pw juveniles, therefore A_3D_3 has prospects of application and promotion on the diagnosis for early stage and present active infection of Paragommiasis (Pw).
