卫氏并殖吸虫感染犬循环抗原的动态观察 被引量:3

KINETICS OF CIRCULATING ANTIGEN BEFORE AND AFTER PRAZIQUANTEL TREATMENT IN DOGS INFECTED WITH PARAGONIMUS WESTERMANI

摘要 应用Dot-ELISA,以抗卫氏并殖吸虫囊蚴和成虫McAbG_2B_3、A_3D_3检测了4只Pw囊蚴感染犬吡喹酮治疗前后血清CAg动态。感染后第4d,4只犬囊蚴CAg均为阳性,1只犬成虫CAg为阳性。至第19d,4只犬囊蚴和成虫CAg均为阳性。第101~105d,4只犬中2只以每只3.2g总量吡喹酮进行治疗,治疗后短期内囊蚴和成虫CAg滴度明显升高,其中尤以成虫CAg升高显著。至治疗后第6d,囊蚴和成虫CAg达高峰,其后便逐步下降。1号犬在治疗后55d、2号犬于治疗后26d囊蚴CAg滴度为0.1号犬于治疗后65d、2号犬于治疗后36d成虫CAg滴度为0。未治疗犬成虫CAg维持于1:16~1:640,囊蚴CAg为1:20~1:40,CAg的动态呈现明显的期特异性。治疗后80d剖检2只治疗犬,其肺部均有多个陈旧性虫囊,虫囊内多为变性坏死虫卵,未发现成虫。未治疗犬于死后即行解剖检获Pw成虫60条。上述研究结果提示:1.感染Pw犬囊蚴CAg的动态具有明显的期特异性,与虫体在宿主体内的发育情况是一致的;2.有效治疗后囊蚴和成虫尤其是成虫CAg的滴度在短期内明显升高;3.可联合或单独应用抗囊蚴McAbG_2B_3、抗成虫McAbA_3D_3进行并殖吸虫病的虫期诊断和疗效考核。 The observation on kinetics of circulating antigen (CAg) before and after Prazi- quantel treatment in dogs infected with Paragonimus westermani was conducted with McAb to metacercaria G_2B_3,McAb to adult worm A_3D_3 by Dot-ELISA. Metacercaria CAg (MCAg)of all 4 dogs (dog No. 1,2,3,4)and adult worm CAg (A CAg) of 1 dog (dog No. 4)were positive 4d after infection. Both M and A CAg can be detected in all 4 dogs 19d after infection. Dog No. 1 and 2 were treated with 3. 2g Praziquantel total dose during 101-105d after infec- tion. Both M and A CAg,especially ACAg titres were distinctly increased shortly after effective treatment with a peak appearing 5-6d post treatment, they began to decline thereafter. The titres of MCAg became 0 55d (dog No. 1) and 26d (dog No. 2) after treatment, so did that of ACAg 65d (dog No. 1)and 36d (dog No. 2)respectively post treatment. No worm was found in treated dogs,while 60 alive adult worms were harvested in the cysts of lungs from infected and untreated dog No. 4 137d after infection. The main points resulting from this observation are as follows: 1. The kinetics of MCAg in dogs infected with P. w were stage specific, coinciding with the development of worm in the hosts. 2. Effective treatment can affect the kinetics of M and A CAg, both M and A CAg titres rais distinctly after effective treatment. 3. McAb G_2B_3 and A_3D_3 might be applied singly or in combination for parasite stage diagnosis of paragonimiasis and efficacy evaluation of treatment.
