摘要
目的 研究高血压左室肥厚时心肌细胞间黏附分子 - 1(ICAM - 1)的表达及其作用。方法 8周龄雄性Wistar-Kyoto (WKY)大鼠和自发性高血压大鼠 (SHR)共 30只 ,分为对照组、高血压组Ⅰ、高血压组Ⅱ ,分别在 2 4、 12、 2 4周龄处死大鼠 ,留取心肌标本进行苏木素 -伊红 (HE)和VanGieson (VG)染色 ,观察心肌细胞形态和胶原分布情况 ;免疫组化染色及ED1标记显示心肌巨噬细胞浸润 ;逆转录 -聚合酶链反应 (RT -PCR)检测心肌ICAM - 1的mRNA表达 ;酶链免疫吸附实验 (ELISA)检测ICAM - 1的蛋白表达。结果 与WKY大鼠及 12周龄SHR比较 ,2 4周龄SHR存在明显的心肌细胞肥大和间质纤维化 ,心肌ICAM - 1的mRNA和蛋白表达水平及巨噬细胞浸润显著增加 (P <0 0 1,P <0 0 1,P <0 0 1) ;12周龄SHR与WKY大鼠在心肌病理学改变、ICAM - 1表达及巨噬细胞浸润等方面无明显差异。结论 心肌ICAM-
Objective To study the expression of myocardial intercellular adhesion molecule-1(ICAM-1) in hypertensive left ventricular hypertrophy.Methods 30 male 8-week-old spontaneously hypertensive rats(SHR) and Wistar-Kyoto(WKY) rats were divided into control group,hypertension groupⅠand hypertension groupⅡ,which were killed at 24,12,24 weeks,respectively. Cardiomyocyte morphology and collagen distribution were observed with HE and Van Gieson(VG)staining. The degree of macrophages infiltration was examined with immunocytochemical staining and ED1 labelling,and the expression of ICAM-1 mRNA and protein in the myocardium was measured with reverse transcription-polymerase chain reaction(RT-PCR) and enzyme-linked immunosorbent assay(ELISA),respectively. Results Compared with WKY rats and 12-week-old SHR,24-week-old SHR showed significant cardiomyocyte hypertrophy,interstitial fibrosis with higher level of ICAM-1 mRNA and protein expression,and greater amount of macrophages infiltration (P<0.01,P<0.01,P<0.01). The pathologic changes,expression of ICAM-1 and the amount of macrophage in myocardium were similar between 12-week-old SHR and WKY rats. Conclusion The macrophage infiltration caused by excessive expression of myocardial ICAM-1 may be involved in genesis of hypertensive left ventricular hypertrophy.
