摘要
顺铂(Cisplatin)是一种抗癌谱广、作用强、与多种抗癌药具有协同作用的抗癌新药。在研究其在肿瘤病人体内的药代动力学时发现:用高效液相色谱法测定家兔血浆药物浓度时,以氯化镍衍生物作内标物尚可,但用于肿瘤病人时,氯化镍衍生物峰变异大,灵敏度低。同时发现病人(或正常人)血中有一物质,其衍生物峰(不衍生化不产生峰)相当稳定,并且随血液的稀释会相应改变其浓度。将它用作顺铂血药浓度测定的内标物,可省去外加内标物的步骤,避免因病人给药后的补液或饮水引起血液的稀释而造成的测量误差,而且还可提高测定方法的准确度。经定性鉴别,血中这一固有物是二价铜元素Cu^(2+)。
By using Cu2+ in human blood as internal standard, determination of cisplatin in patients plasma has been performed by CFD-RPHPLC. Chromatographic conditions were : YWG-C18 H37· ψ4 × 250mm column, methanol-water (4 : 1)as mobile phase and UV detection at 254nm. Correlation coefficient was 0. 9921 and the average recoveries for samples were 102. 6% ±6. 7%.
